A ready-made scale

Let one square equal one inch of finished quilt and a Letter sheet covers about 16 by 21 inches at half-inch squares, a single block at generous size, or a full throw if you let one square equal two inches. Because the print is true size. You can lay a rotary ruler straight on the sheet and measure.

Half inch for blocks, quarter inch for detail

Half-inch squares suit block layout and quilt-top planning: big enough to write in, few enough to count at a glance. Drop to quarter-inch when you are working out the piecing inside a single complex block and need the finer resolution. Both print at true size, so you can move between them without rescaling anything.

Seam allowance is not on the paper

Quilt blocks are drawn at finished size; the quarter-inch seam allowance is added when you cut. Drawing allowances into the plan is a common early mistake and makes every block finish small. Plan finished, cut with allowance. Worth knowing too that experienced quilters usually sew what is called a scant quarter inch, a thread or two narrower than a true quarter, because the fold of the pressed seam itself consumes a little fabric. Over a block with many seams that difference accumulates into a block that finishes measurably small. The paper cannot help with this, but it is the usual explanation when blocks planned correctly still come out undersized.

Match the grid to the piecing

Squared paper handles rectangular piecing well and nothing else. For half-square and equilateral triangles use the triangle grid; for English paper piecing and hexie quilts use hexagon paper; for tumbling blocks and other 3D illusions use isometric paper, where the diamond is already drawn for you. Planning 60° shapes on square paper is the single most common reason a pieced design does not come out as drawn.

Printing it accurately

This only measures correctly if your printer doesn't rescale the page. In the print dialog set Scale: 100%, not "Fit to page" or "Shrink oversized pages". If you're not sure it worked, print the calibration page, measure the ruler on it with a real ruler, and it will tell you exactly what your printer is doing to the page.
