One square, one intersection
Needlepoint is worked over the intersections of a woven canvas, and that mesh is genuinely square: the stitches are as tall as they are wide. So the chart paper is square too, with each cell standing for one intersection rather than one thread. Charting a needlepoint design on knitting paper, which is deliberately wider than it is tall, will squash the design in a way that only becomes obvious once it is stitched.
Mesh count sets the finished size
Canvas is sold by mesh — 10, 12, 13, 14 and 18 holes to the inch are the common ones. Divide your chart's stitch count by the mesh to get the finished dimension: a 140-stitch design comes out at 10 inches on 14 mesh and 7.8 inches on 18. Decide the mesh before you chart rather than after, because the same chart produces quite different objects.
Charting stitches that cover more than one hole
Basketweave, continental and half-cross all cover a single intersection and chart identically, one cell each. Decorative stitches — Scotch, mosaic, Byzantine — cover blocks of two, three or four intersections and have to be charted as the block they occupy. Outline those areas on the chart first, then fill the plain tent stitch around them.
Printing it accurately
This only measures correctly if your printer doesn't rescale the page. In the print dialog set Scale: 100% — not "Fit to page" or "Shrink oversized pages". If you're not sure it worked, print the calibration page, measure the ruler on it with a real ruler, and it will tell you exactly what your printer is doing to the page.
